《洗冤录》着于宋代，是法医史上的惊世巨著，其作者却是一个饱受歧视的棺材遗腹子。30岁以前，他一直靠打更为生，不料一宗命案改变了他的人生……因分地纠纷而找村长理论的宋慈与劫富济贫的江湖女贼唐思意外地卷入了谋杀村长的案中，被判死刑，后得到新知县宋翊及熟悉验尸的马贵相助，沉冤得雪。宋翊聘宋慈为忤作，两人合作屡破奇案，声名大噪。唐思因宋翊救命之恩，暗生情愫，但宋翊已与聂枫发展感情。这时一名长相与宋翊一模一样的人突然出现，原来假宋翊为求功名，将真宋翊推下山崖冒名顶替，做起了知县；后又攀附权贵，抛弃聂枫而娶富家女蓝彩蝶为妻，岂料蓝家早已外强中干，负债累累。假宋翊感到彩蝶是他的负担，故设计陷害。其奸险面目渐渐表露，但他每次行凶时均非常谨慎，都要彻底毁尸灭迹。宋慈无计可施之际，意外地发现了真宋翊的尸体，经过艰苦地调查取证，在唐思和聂枫的帮助下，最终惩戒了凶手，伸张了正义。
